Identification of a candidate gene responsible for male sterility conferred by CMS-T cytoplasm in onion (Allium cepa L.) and development of molecular markers for detection of CMS-T cytoplasm

Cytoplasmic male sterility (CMS) has been exclusively used as an emasculation tool in seed production of hybrids onion (Allium cepa L.).Three types CMS (CMS-S, CMS-R, and CMS-T) have hybrid breeding. Male conferred by both CMS-S CMS-R is likely induced open reading frame (orf) 725, a chimeric gene. However, this gene was not detected accessions containing CMS-T cytoplasm. Instead, region showing high homology with orfA501, which located the 3’ end orf725, CMS-T. A novel gene, we named orf219, identified genome walking PCR amplification based on orfA501 homolog. The 684-bp orf219 consisted 128-bp exon 1 atp1 556-bp sequence high-copy-number only cytoplasm (T cytotype). Analysis RT-PCR products showed normal transcription eight RNA editing sites, one created stop codon, resulting shorter amino acid upon translation. Sequences four hypervariable regions chloroplast organization syntenic blocks mitochondrial indicated that very closely related to (N) male-fertile cytotypes. Based these findings, new molecular markers were developed for identification cytotypes CMS-T, -R, -S, -N. Cytotypes 424 diverse using six markers. predominated among analyzed accessions.
